CRISPR/Cas9 technology enables the rapid generation of loss-of-function mutations in a targeted gene in mammalian cells. CRISPR-Cas9 knock-out cell lines from Cellalabs provide you with reliable, off-the-shelf, single gene knock-outs in biologically relevant cell lines for target validation, pathway discovery, and more, making it easy to interrogate your target of interest. According to the characteristics of different cell lines, appropriate methods will be selected to get the positive clones.

The standard workflow to generate CRISPR KO cell lines involves five major steps, 

  1. Choosing a knockout strategy. 
  2. Selecting gRNA target sites and performing vector cloning.
  3. Introducing gRNAs by transfection or transduction.
  4. Isolation and expansion of single-cell clones.
  5. Knockout verification by western blot analysis, PCR, and/or Sanger sequencing.

Knock-in Cell Line

Targeted genome editing events fall into two general classes: knock-in and knock-out. Knock-in mutations typically require an exact DNA sequence to be incorporated at a precise genomic location. A Knock-in cell line defines a cell line in which an exogenous gene is inserted in a native locus of interest. Identification of gene functions in cell line models is important for gene discovery and exploration of cell-intrinsic mechanisms. The gRNA vector (containing Cas9) of the CRISPR/Cas9 system and the Donor DNA carrying target mutation or target gene sequences will be co-transfected into cells by electroporation. Through antibiotics selection, the transfected cells were successfully got. For further selection, the single-positive clone was validated by PCR and Sanger sequencing.
